Why Science? | ICH M9 BCS Biowaivers, Solubility and Permeability

Three students sit around open books and worksheets at a classroom table, reading, writing and discussing the work together.

eduKateSG · Why Science?

Use solubility, intestinal permeability, formulation and dissolution evidence to decide when an immediate-release oral product may be compared without a new in-vivo bioequivalence study

Read FDA’s final May 2021 ICH M9 guidance and May 2023 questions and answers through BCS classes, dose solubility, permeability evidence, excipient similarity, dissolution profiles, eligibility, exclusions and dossier documentation.

Full chapter index · Science Learning Hub

Bioequivalence usually asks whether two products deliver a drug into the body at a comparable rate and extent. For some immediate-release oral products, carefully designed laboratory evidence can answer enough of that question to waive an additional human bioequivalence study.

FDA lists ICH M9 as final Level 2 Guidance dated May 2021. The official page also links final questions and answers from May 2023 that clarify implementation.

Did you know? The Biopharmaceutics Classification System combines two drug-substance properties: aqueous solubility and intestinal permeability. Product composition and dissolution then determine whether a particular dosage form qualifies for a waiver.

A BCS biowaiver is not a shortcut around science. It replaces one kind of evidence with a tightly bounded package of physicochemical, biological and comparative product evidence.
