Estimate viable aerobic microorganisms in a non-sterile product while proving that the product matrix does not hide them
Read FDA’s final April 2009 Q4B Annex 4A through total aerobic microbial count, total yeast and mould count, sampling, media, dilution, neutralisation, method suitability, plating, incubation, counting, acceptance and investigations.
Non-sterile does not mean microbiologically uncontrolled. Tablets, creams, liquids and raw materials can allow limited microbial presence, but excessive or unsuitable contamination may signal a process problem or create risk.
FDA lists Q4B Annex 4A, Microbiological Examination of Non-Sterile Products: Microbial Enumeration Tests General Chapter, as final guidance dated April 2009 and notes an FDA page update on 18 September 2017.
Did you know? A preservative that protects a product can also suppress organisms during the laboratory test. Method suitability therefore asks whether the procedure can recover microorganisms in the presence of the product matrix.
For students, enumeration testing brings microbiology to life: one visible colony may represent a cell or cluster that survived sampling, dilution, recovery and incubation. Every step shapes what can be counted.
